@EN Thyroid cell culture is very important for thyroid research. Currently wide varieties of animal thyroid and human thyroid are using for thyroid cell culture. We cultured the porcine thyroid cells and studied the thyroglobulin synthesis and
secretion. And also we studied the hormonal effect of the thyroglobulin synthesis and secretion from the mono-layer cultured porcine thyroid cells.
Primary thyroid follicular epithelial cells isolated from fresh porcine thyroid tissue by sequential trypsinization in the presence of EGTA were washed twice in DMEM containing 10% newborn calf serum (NCS) at the end of digestion to quench
residual
trypsin. Cells were than seeded at high density (1¡¿10E6 cells/§²) in DMEM containing 1% NCS onto 24-well plates which were precoated with matrigel. Next day, medias were changed for serum-free DMEM with different hormone conditions.
The growth of thyroid cells and the thyroglobulin formation from the cultured thyroid cells were excellent, and the synthesis of the thyroglobulin and cell proliferation were regulated by the hormone conditions in the media.
In conclusion, cultured porcine thyroid cells were very useful in thyroid and thyroglobulin research, and hormone conditions in medial were considered as important regulating factor of thyroid cell growth-an thyroglobulin-associated research (J
